Addgene recommends the following steps to ensure proper handling and storage of your pooled library. If there is a problem, please contact Addgene within 30 days of receiving your pooled library.
Pooled Library Storage
Most libraries from Addgene arrive as a pooled DNA library on blue ice. It may not be frozen. Addgene recommends storage at 4°C. For long-term storage, Addgene recommends storage at -20°C. If you choose to store the library at -20°C, we strongly recommend minimizing the number of freeze/thaw cycles by aliquoting the sample.
A few libraries are shipped on dry ice. These should arrive frozen on dry ice and should be immediately stored -80°C.
Pooled Library Concentration
The volume and concentration of DNA you receive will depend upon which library you ordered. See your library's Addgene web page for details.
Addgene has measured the concentration of the library you received, and this value is indicated on the library page. The DNA can be used directly for amplification according to the concentration on our website. We do not recommend using any of the sample for measuring concentration, as that volume represents DNA that will be lost and unable to contribute to a successful amplification.
Pooled Library Recovery
To access the library, we recommend you spin down the tube before opening the lid. In most cases, you will need to amplify the pooled library you received from Addgene before using it. Please find any amplification or other protocols for the specific library you ordered on its Addgene product page.
Please note that we recommend following protocols exactly as described by the depositing lab. Use of alternative protocols/reagents could result in loss of representation or recombination of large library backbones, such as lentiviral vectors.
Representation of amplified gRNA libraries must be verified by next generation sequencing prior to use in a screen. Click here for more information on pooled library diversity. Information on pooled library quality control can be found here. When performing NGS on an amplified library sample, we recommend sequencing a pre-amplification control in parallel to the amplified sample. Please notify us within 30 days of receipt if there are problems detected in the pre-amplification control.

Any and all information Addgene has about your library can be found on Addgene’s website. Search using the library name or ID number listed on your Order Summary. Alternatively, you can use the links from your Order Confirmation email.
Pooled Library Verification
Addgene cannot guarantee that the depositing laboratories have submitted the correct material. Please notify Addgene of any problems within 30 days.
Please visit www.addgene.org and search the catalog number of your pooled library for any recommended protocols from the depositor.
Citations in Future Publications
The pooled library you have received was created by your colleagues. Please acknowledge the Principal Investigator, cite the article listed on the pooled library datasheet, and include the Addgene catalog ID in future publications.
